Reports of histone-activated SIR2-mediated NAD+ metabolism and NAD+-activated SIR2-mediated histone deacetylation suggest a coupled reciprocal activation mechanism involving interactions of SIR2 with NAD+ and the N-ε-acetyl-lysine groups of acetylated histone.
