  • TNF-α converting enzyme (TACE, also known as ADAM17) is a zinc metalloprotease that cleaves the membrane-bound precursor of TNF-α to give the mature soluble form of TNF-α.{24091,24093} It also mediates the proteolytic release of several other cell-surface proteins, including TNF-α receptors (TNFRs). TAPI-1, known in the earlier literature as just TAPI, is an inhibitor of TACE that inhibits the cleavage of TNF-α, TNFRI (p60), and TNFRII (p80) with IC50 values ranging from 5-100 µM.{24091,29961,24092} It also blocks the shedding of other TACE-dependent proteins, including IL-6R and Klotho, and may inhibit metalloproteases other than TACE.{24093,15600,29962} TAPI-1 differs from TAPI-0 (Item No. 14694), structurally, by the presence of an aminoethyl addition at the alanyl group, and is generally more stable in tissue culture and in vivo.

  • Tau is a member of the microtubule-associated protein (MAP) family that facilitates microtubule formation in axons.{55125,55126} It is primarily expressed in neurons and localized to axons, but trace amounts have been observed in glial cells and several peripheral tissues including kidney, lung, and testis. Tau is composed of an N-terminal projection domain that interacts with neurofilaments, cytoplasmic organelles, and neuronal cell membranes and a C-terminal microtubule binding domain that facilitates microtubule polymerization and stabilization. It is encoded by MAPT in humans, a 16-exon gene that produces 6 isoforms via alternative mRNA splicing. Mutations in MAPT lead to changes in expression of tau isoforms and the formation of insoluble protein aggregates that cause familial frontotemporal dementia (FTD) and parkinsonism linked to chromosome 17 (FTDP-17).{55127} Tau is subject to post-translational modifications, including phosphorylation, and hyperphosphorylation of tau is associated with the formation of neurofibrillary tangles and neuronal cell death in postmortem brains from patients with Alzheimer’s disease. Cayman’s Tau Polyclonal Antibody can be used for immunohistochemistry (IHC), immunocytochemistry (ICC), and Western blot (WB) applications. The antibody recognizes three isoforms of tau at approximately 48, 65, and 75 kDa from human, bovine, mouse, porcine, and rat samples.

The concentration was verified by comparison to an independently prepared calibration standard. This TCDCA MaxSpec® standard is guaranteed to meet identity, purity, stability, and concentration specifications and is provided with a batch-specific certificate of analysis. Ongoing stability testing is performed to ensure the concentration remains accurate throughout the shelf life of the product. Note: The amount of solution added to the vial is in excess of the listed amount. Therefore, it is necessary to accurately measure volumes for preparation of calibration standards. Follow recommended storage and handling conditions to maintain product quality.

The solution has been prepared gravimetrically and is supplied in a deactivated glass ampule sealed under argon. The concentration was verified by comparison to an independently prepared calibration standard. This TUDCA MaxSpec® standard is guaranteed to meet identity, purity, stability, and concentration specifications and is provided with a batch-specific certificate of analysis.

  • Lipid peroxidation is a well-established mechanism of cellular injury in both plants and animals and is used as an indicator of oxidative stress in cells and tissues. Lipid peroxides, derived from PUFAs, are unstable and decompose to form a complex series of compounds, which include reactive carbonyl compounds, such as malondialdehyde (MDA). MDA can be quantified through a controlled reaction with thiobarbituric acid, generating ‘Thiobarbituric Acid Reactive Substances’ (TBARS). Cayman’s TBARS Assay Kit provides a simple, reproducible, and standardized tool for assaying lipid peroxidation in plasma, serum, urine, tissue homogenates, and cell lysates. The MDA-TBA adduct formed by the reaction of MDA and TBA under high temperature (90-100°C) and acidic conditions can be measured either colorimetrically at 530-540 nm or with much higher sensitivity fluorometrically at an excitation wavelength of 530 nm and an emission wavelength of 550 nm.

  • Oxidative stress in the cellular environment results in the formation of highly reactive and unstable lipid hydroperoxides. Decomposition of the unstable peroxides derived from PUFAs results in the formation of malondialdehyde (MDA), which can be quantified colorimetrically following its controlled reaction with thiobarbituric acid. The measurement of these ‘Thiobarbituric Acid Reactive Substances’ (TBARS) is a well-established method for screening and monitoring lipid peroxidation.{14303,14304} Modifications of the TBARS assay has been used to evaluate a wide range of samples that include human and animal tissues and fluids, drugs and foods. Even though there remains a controversy cited in literature regarding the specificity of TBARS toward compounds other than MDA, it still remains the most widely employed assay used to determine lipid peroxidation. Cayman’s TBARS Assay Kit provides a simple, reproducible, and standardized tool for assaying lipid peroxidation in plasma, serum, urine, tissue homogenates, and cell lysates.

  • TBB is an ATP/GTP-competitive inhibitor of casein kinase 2 (CK2; IC50 = 0.15 µM for rat liver CK2).{22897} It also inhibits PIM3 and DYRK2 with IC50 values of 0.86 and 0.99 µM, respectively, and several other protein kinases with IC50 values in the low micromolar range but displays less than 10% inhibition at 26 additional kinases up to 10 µM.{22896,22897}

  • Casein kinase 2 (CK2) is a kinase with many targets and diverse roles in cell signaling and disease.{26170,26171} TBCA is a selective, cell-permeable inhibitor of CK2 (IC50 = 0.11 µM, Ki = 77 nM) that has minimal effect on a panel of 28 other kinases.{24973} It induces cell death in Jurkat adult T cell leukemia cells (DC50 = 7.7 µM), driving caspase-dependent degradation of poly-ADP ribose polymerase while inhibiting CK2 activity.{24973} TBCA also supports a role for CK2 in platelet function and Akt phosphorylation but not protein splicing.{26172,26173}
